Charlotte Christiana Sophia wurde als dritte Tochter Ludwig Rudolphs und seiner Gemahlin Christine Luise von Oettingen-Oettingen am 28. August 1694 geboren. Sie wurde am 25. Oktober 1711 mit dem Zarewitsch Alexei von Rußland, dem Sohn des Zaren Peter I., verheiratet. Das Paar hatte zwei Kinder: Tochter Natalija (1714-1728) und den später regierenden Zar Peter II. (reg. 1727-1730). Kurz nach der Geburt Peters starb Charlotte am 1. November 1715 an Kindbettfieber.
